The presence of angiotensin II receptors was investigated in renal endocrine cells by light and electron microscopic radioautography and by double immunolabeling of intracellular peptides and receptors. Angiotensin II type 1 receptors were demonstrated in all the typical endocrine cells of the pars intermedia of the rat adrenal gland, the proximal tubules of the kidney, and the chromaffin cells of the adrenal medulla. In the pars intermedia of the rat adrenal gland, a large majority of the receptors were localized in cells of the zona glomerulosa and fasciculata, the cells that were identified by immunocytochemistry to contain the antidiuretic hormone (ADH) or corticotropin-releasing hormone (CRH). Electron microscopy and double immunolabeling revealed that the angiotensin II type 1 receptors in the cells of the zona glomerulosa were adjacent to intramembrane particles which contained the antigenic determinants of ADH. The pars intermedia of the rat adrenal gland was also intensely labeled for angiotensin II type 1 receptors; the highest densities of receptors were localized in cells located at the innermost layer of the zona fasciculata. In the pars intermedia of the rat adrenal gland, angiotensin II type 1 receptors were mainly localized in neurosecretory granules. In the renal proximal tubules, angiotensin II type 1 receptors were observed in cells with secretory granules and zymogen granules. Double immunolabeling indicated that at least some of the receptor-containing granules were positive for ADH, renin, and aspartate aminotransferase, antigens typical of the granules of these cells. In the chromaffin cells of the adrenal medulla, a few angiotensin II type 1 receptors were observed in an association with secretory granules. These studies indicate that angiotensin II is involved in the control of several functions of endocrine cells in the rat kidney and the adrenal gland and that the endocrine cells may contain more than one peptide receptor mediating
